I think the best italian food in soho is a place called Da Mikele 275 Church St (between Avenue Of The Americas & White St) New York, NY 10013 It's actually considered Tribeca but worth the few minutes walk from anywhere in Soho. "old-fashioned square pies like the Diavola, which is topped with San Marzano tomatoes and spicy salame, or the patate, a white pie that features potatoes, spec, rosemary and garlic. There's a large selection of other Italian specialties, including homemade foccacia bread, oven toasted flat breads, and pasta options like the paccheri with shrimp, white beans and calamari."

If your looking for a more upscale Italian food experience in Soho check out the new Isola Trattoria At The Mondrian Soho: 9 Crosby St, btwn Grand and Howard St; Soho. Set up like the fanciest Italian back garden you've ever seen, complete with lush trees, chandelier clusters, an overhead centerpiece adorned with glass vases, and marble-topped tables held up by Roman columns. Food items include Handmade pasta (bucatini w/ fennel sausage, crab & Calabrese peppers) and pizza (burrata, squash blossom pesto, and roasted tomato) Also non-traditional Italian like the Isola Burger w/ truffled lardo, balsamic red onions & gem lettuce.
